S100B蛋白在C57/BL6J小鼠螺旋神经节细胞内的表达

摘要
目的研究S100B蛋白在C57/BL6J小鼠螺旋神经节细胞内的表达,探讨其在老年性聋发病中的作用。方法将40只C57/BL6J小鼠分成2月龄和12月龄组,每组20只,完成ABR检测后,取每只小鼠内眦静脉血0.8ml,采用ELISA法检测小鼠血清中S100B的含量,HE染色观察小鼠耳蜗螺旋神经节细胞形态,免疫组化检测螺旋神经节细胞内S100B蛋白表达。结果 2、12月龄组小鼠平均ABR阈值分别为20±5、55±5dB SPL,前者低于后者(P<0.05);血清中S100B含量分别为0.255±0.032、0.869±0.045μl/g,前者低于后者(P<0.05);S100B蛋白在两组小鼠螺旋神经节细胞均有表达,表达部位和密度不同,2月龄小鼠主要分布在胞膜和胞浆,12月龄小鼠则胞核见大量表达,2、12月龄小鼠S100B表达光密度值(IOD)分别为1.106±0.030、3.558±0.05,前者低于后者(P<0.05)。结论 12月龄C57/BL6J小鼠听功能符合老年性聋改变,其体内血清中S100B含量比2月龄小鼠增高,耳蜗螺旋神经节细胞内S100B蛋白表达较2月龄小鼠增多,推测S100B蛋白可能参与老年性聋的发病过程。
