siRNA抑制GRP75表达对叔丁基双氧水诱导HEI-OC1毛细胞株凋亡的影响

摘要
目的分析抑制热休克蛋白75(GRP75)对氧化应激损伤HEI-OC1细胞凋亡的影响,探讨GRP75在耳蜗毛细胞凋亡过程中的作用机制。方法人工设计合成靶向GRP75的小干扰RNA(small interference RNA,siRNA),转染不同浓度(20、40、60μM)叔丁基过氧化氢(t-BHP)染毒HEI-OC1毛细胞株,并设空白对照,采用蛋白免疫印迹法检测GRP75和Caspase-3蛋白表达水平。结果转染GRP75-siRNA后HEI-OC1细胞GRP75表达水平明显受到抑制(t=35.722,P<0.01);T-BHP染毒组GRP75表达水平明显升高(细胞凋亡组:F=1 009.09,P<0.01;siRNA转染组:F=1 603.97,P<0.01),并且都随着染毒浓度的增加而增加;T-BHP染毒组Caspase-3表达水平显著增高(细胞凋亡组:F=3 068.67,P<0.01;siRNA转染组:F=6 348.27,P<0.01),并都随着染毒浓度的增加而增加;20μM、40μM、60μM t-BHP染毒组转染siRNA后Caspase-3表达水平均显著升高(t=21.275、P<0.01;t=18.584、P<0.01;t=11.147、P<0.01)。结论抑制氧化应激损伤毛细胞的GRP75表达可导致凋亡程度加重,推测GRP75可能具有抗凋亡效应。
