慢性睡眠剥夺和高脂饮食与咽喉反流性疾病相关性的实验研究

摘要
目的探讨慢性睡眠剥夺和高脂饮食与咽喉反流性疾病(laryngo pharyngeal reflux disease,LPRD)的相关性。方法 16只大鼠随机分为实验组和对照组,每组8只,实验组采用改良多平台水平面法剥夺大鼠白天睡眠,每日8小时,同时喂食含35%脂肪的高脂饲料,对照组自由睡眠和喂食普通饲料,2个月后使用双探头上消化道pH值监测仪连续10小时检测两组大鼠咽部及食管pH值,比较两组大鼠实际检测的pH<4的总时间百分率(反流指数)、pH<4的总反流次数(换算成24小时数值)和最长反流持续时间(min)的差异。结果模型构建过程中无大鼠死亡,实验组与对照组大鼠体重增加差异无统计学意义(P>0.05);实验组大鼠咽部反流指数、最长反流时间和24小时总反流次数中位数分别为21.07、27.25和61.11,对照组分别为1.25、0.95和8.44,实验组三项指标均较对照组明显增多或延长(P<0.05),提示实验组大鼠出现了咽喉反流性疾病;实验组食管反流指数和24小时总反流次数中位数分别为15.87和96.67,对照组分别为2.58和13.27,实验组较对照组明显增多(P<0.05);实验组和对照组食管最长反流时间中位数分别为26.25和5.6,实验组比对照组延长,但差异无统计学意义(P>0.05)。结论慢性睡眠剥夺和高脂饮食可导致大鼠发生LPRD,该法可能成为新的LPRD动物模型造模方法。
