醋酸去氨加压素对豚鼠耳蜗水通道蛋白2表达的调节作用

摘要
目的观察醋酸去氨加压素(deamino arginine vasopressin,DDAVP)对内淋巴积水模型豚鼠耳蜗水通道蛋白2(aquaporins 2,AQP2)表达的调节,探讨梅尼埃病的发病机制。方法 40只成年豚鼠随机分为实验组和对照组,每组各20只。实验组豚鼠腹腔注射DDAVP 4μg·kg-1·d-1,共7天,构建内淋巴积水模型,对照组豚鼠腹腔注射等量生理盐水。停药7天后分别取两组各5只豚鼠耳蜗切片行HE染色,计算中阶与中阶加前庭阶横截面积比值平均值(R值)评估内淋巴积水程度,同时,2组各15只豚鼠分别采用免疫组化法和蛋白印迹法检测耳蜗AQP2蛋白的表达。结果实验组和对照组的R值分别为0.42±0.04和0.31±0.03,实验组豚鼠内淋巴积水程度重于对照组(P<0.05);实验组和对照组豚鼠耳蜗AQP2均表达于血管纹、螺旋神经节细胞,其中,实验组耳蜗AQP2蛋白半定量分析(0.9954±0.0498)高于对照组(0.7664±0.1452),差异有统计学意义(P<0.05)。结论DDAVP上调豚鼠耳蜗中与水转运密切相关的AQP2的基因表达,参与内淋巴积水形成。
