单侧耳蜗损毁后小鼠耳蜗核及下丘生长相关蛋白-43的表达

The Expression of GAP-43 in Cochlear Nucleus and Inferior Colliculus after Unilateral Cochlear Damage in Rats

目的研究小鼠单侧耳蜗损毁后不同时间点耳蜗核(cochlear nucleus,CN)和下丘(inferior colliculus,IC)生长相关蛋白-43(growth associated protein-43,GAP-43)的表达变化,探讨小鼠脑干听觉中枢的可塑性。方法将成年健康小鼠30只随机分为耳蜗损毁术后3、7、15、30、60天组及对照组,每组5只。前5组制成单侧耳蜗损伤模型,对照组仅行耳后切口、缝合,不损毁耳蜗。采用免疫组化SP法检测各组耳蜗核和下丘GAP-43蛋白的表达变化。结果①小鼠单侧耳蜗损毁后3、7、15天,术侧耳蜗核GAP-43较对照组表达升高(P<0.05);术后30天,术侧GAP-43表达水平下降,但仍高于对照组及对侧(均P<0.05);术后60天,术侧GAP-43表达水平与对照组差异无统计学意义(P>0.05);对侧耳耳蜗核GAP-43含量水平与对照组差异均无统计学意义(P>0.05)。②小鼠单侧耳蜗损毁后3、7、15天,术耳同侧和对侧下丘GAP-43表达水平较对照组升高,且表达均持续上升,差异有统计学意义(P<0.05),术耳同侧表达水平低于对侧(P<0.05);术后30天表达逐渐下降,但术耳同侧和对侧仍高于正常对照组(P<0.05),且术侧低于对侧(P<0.05);术后60天其表达继续下降,术耳同侧及对侧与对照组、术耳同侧与对侧表达差异均无统计学意义(均P>0.05)。结论单侧耳蜗损毁后,小鼠耳蜗核和下丘核GAP-43的表达先升高后下降,可能反映了脑干听觉中枢神经元在听力损失后轴突的再生及突触重塑。
