分泌性中耳炎模型大鼠中耳黏膜中上皮钠通道mRNA的表达及其意义

摘要
目的探讨上皮钠通道(epithelial sodiumchannel,ENaC)mRNA在分泌性中耳炎模型大鼠中耳黏膜中的表达及其意义。方法 12只SD大鼠实验耳经鼓膜注入脂多糖(lipopolysaccharide,LPS)制造分泌性中耳炎(otitis media with effusion,OME)动物模型,在造模后第二天取中耳黏膜,采用实时定量-聚合酶链反应(real time-polymerase chain reaction,Red ti me PCR)检测中耳黏膜中ENaC-mRNA的表达,并与对照耳(注入等量生理盐水)相比较。结果实验耳α-、β-、γ-ENaC-mRNA的相对表达量分别为0.007 8、0.009 2、0.007 6,对照耳α-、β-、γ-ENaC-mRNA的相对表达量分别为0.012 1、0.012 6、0.011 7;实验耳相α-、β-、γ-ENaC-mRNA相对于对照组分别下调1.56倍(P<0.05)、1.37倍(P<0.05)、1.53倍(P<0.01)。结论 ENaC在分泌性中耳炎大鼠模型中耳黏膜中基因转录水平发生下调,这可能是导致OME早期积液形成的机制之一。
