缺氧诱导因子在分泌性中耳炎大鼠模型中耳黏膜中的表达

摘要
目的建立分泌性中耳炎动物模型并检测缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)在中耳黏膜中的表达。方法取34只排除中耳病变的SD大鼠,从软腭进路以三氯醋酸烧灼一侧咽鼓管咽口建立分泌性中耳炎模型(实验组),另一侧耳为对照组,采用免疫组化与逆转录-聚合酶链反应(RT-PCR)技术检测HIF-1α在实验组和对照组中耳黏膜的表达。结果分泌性中耳炎动物模型建模成功14例(14耳),建模平均时间为1周;正常对照组中耳黏膜HIF-1αmRNA表达灰度值为5.297±0.697,实验组为6.870±0.632,两组间差异有统计学意义(P<0.05);正常对照组HIF-1α蛋白水平免疫组化染色计分为0.500±0.149,实验组染色计分为1.530±0.337,两组间差异有统计学意义(P<0.05)。结论采用化学烧灼法烧灼咽鼓管咽口能模拟咽鼓管功能不良成功建立分泌性中耳炎动物模型,HIF-1α在分泌性中耳炎模型动物中耳黏膜中表达增多。
