强化铁营养对缺铁大鼠耳蜗膜性组织actin、myosin、desmin和HSP表达的影响

Effects of Fortified Iron Nutrition on Actin,Myosin,Desmin,Heat Shock Protein Expression in the Membranous Cochlea in Iron-Deficient Rats

摘要
目的应用蛋白质组学技术研究强化铁营养对缺铁大鼠耳蜗膜性组织肌动蛋白(actin)、肌球蛋白(myosin)、结蛋白(desmin)和热休克蛋白(heat shock protein,HSP)表达的影响。方法体重25~35g、健康、无耳疾、ABR阈值正常的1~2周龄Wistar大鼠120只,随机分成正常对照组20只,喂以标准饲料16周;缺铁大鼠组100只,喂以缺铁饲料8周后,复制出至少有一耳ABR阈值改变≥15dB的大鼠共41只,再将该41只大鼠随机分成缺铁耳聋组20只和强化铁治疗组21只,缺铁耳聋组继续喂以缺铁饲料8周、强化铁治疗组喂以强化铁饲料8周后,取大鼠耳蜗膜性组织进行蛋白质组学分析,对各组间actin、myosin、desmin和HSP表达的差异进行比较。结果缺铁耳聋组大鼠耳蜗膜性组织中的actin、myosin及HSP种类较正常对照组减少,并出现特异蛋白质desmin;强化铁治疗组大鼠耳蜗膜性组织myosin和HSP种类较缺铁耳聋组增加,actin种类未见明显改变,desmin消失。结论铁缺乏导致的耳蜗actin、myosin及HSP种类减少,以及出现特异性蛋白质desmin,可能与缺铁性聋的发生发展有关;强化铁营养可使缺铁耳聋大鼠耳蜗myosin和HSP种类增加,desmin消失。
