电针对硫酸卡那霉素慢性损害后豚鼠听皮层抑制素和吞蛋白-A1表达的影响

The Effects of Electroacupuncture on Prohibition and Endophilin-A1 Expression in Auditory Cortex of Guinea Pig Following Chronic Kanamycin-Induced Damage

摘要
目的 探讨硫酸卡那霉素慢性损害后豚鼠听皮层蛋白质组的变化,以及电针对抑制素和吞蛋白-A1表达的影响。方法 105只成年健康杂色豚鼠随机分为对照组(7只)、硫酸卡那霉素模型组(49只,模型组)、硫酸卡那霉素+电针组(49只,电针组)。模型组与电针组连续注射硫酸卡那霉素7天,电针组注射半小时后予以电针翳风穴和听宫穴15分钟,连续7天,对照组注射等量生理盐水;模型组与电针组根据药物注射后时间再分为注射后1、7、14、28、56、70、140天组(每组7只)。采用双向电泳技术、质谱鉴定分析筛选并切取21个表达差异明显的蛋白点进行鉴定,蛋白质印迹法检测抑制素与吞蛋白-A1的表达。结果 (1)灰度差异2倍以上的差异表达蛋白质有抑制素、吞蛋白-A1等18种。(2)与对照组相比,模型组注射后第14、28、56天抑制素表达降低(P<0.05),第1、14天吞蛋白-A1表达增高(P<0.05);与模型组相比,电针组注射后第7、70天抑制素表达增高(P<0.05),第14、28、56天吞蛋白-A1表达降低(P<0.05)。结论 抑制素等18种蛋白质可能参与了硫酸卡那霉素致豚鼠听皮层慢性损害的过程,电针听宫穴与翳风穴可能通过调节抑制素、吞蛋白-A1的表达促进听皮层的重塑进程。
