PD98059抑制大鼠听泡成骨细胞增殖及分化

PD98059 Inhibits the Proliferation and Differentiation of Osteoblasts in Rat Otocyst

摘要
目的 探究ERK抑制剂PD98059对大鼠听泡成骨细胞增殖及分化的影响。方法 用0.25%胰酶和I型胶原酶两步消化法提取SD乳鼠听泡成骨细胞,分别与浓度为0、10、25和50μmol/L的ERK抑制剂PD98059进行共培养,然后采用EDU法连续4天对4组成骨细胞的增殖进行检测,对各组增殖趋势进行对比分析。用10 mmol/L、50μg/ml和10~(-7) mol/L的浓度的β-甘油磷酸钠、L-维生素C和地塞米松对4组成骨细胞进行成骨诱导分化,24 h后采用RT-qPCR技术检测各组成骨相关因子Ocn、Bsp、Runx2、Bmp2、OPG和RANKL mRNA的表达水平,分析结果。结果 浓度为10、25和50μmol/L的ERK抑制剂PD98059对SD乳鼠听泡成骨细胞的增殖均有不同程度的抑制作用,其中浓度为10μmol/L的PD98059对其增殖的抑制作用明显大于其余3组(P<0.05)。此外,上述不同浓度的PD98059对Ocn、Bsp、Runx2、Bmp2和OPG mRNA的表达均有不同程度的抑制作用,其中Ocn、Bsp、Runx2和Bmp2 mRNA的表达在10μmol/L的PD98059组明显低于0、25和50μmol/L的PD98059组(P<0.05);OPG mRNA的表达在10和25μmol/L的PD98059组明显低于0和50μmol/L的PD98059组(P<0.05),而前二者间的差异并无明显统计学意义(P>0.05)。RANKL mRNA的表达在各组中均未检测出CT值。结论 ERK通路抑制剂PD98059对SD乳鼠听泡成骨细胞的增殖和分化均有抑制作用,推测ERK1/2-MAPK通路可能通过影响大鼠听泡成骨细胞的增殖与分化而介导了鼓室硬化的形成。
