中耳胆脂瘤中活性氧及磷酸化蛋白激酶的表达及相关性研究

摘要
目的检测中耳胆脂瘤上皮组织中活性氧(reactive oxygen species, ROS)、磷酸化蛋白激酶B(phosphorylated protein kinase B,P-AKT)的表达及两者的相关性,探讨ROS在中耳胆脂瘤发病机制中的作用。方法应用免疫荧光技术检测中国医科大学附属盛京医院2019年9月~12月期间25例中耳胆脂瘤标本及15例正常皮肤标本中ROS的表达量;应用Western blot免疫印迹法检测上述标本中P-AKT和内参GAPDH的表达;并进一步分析ROS和P-AKT表达的相关性。结果免疫荧光染色显示,ROS定位于胆脂瘤和正常皮肤的细胞核,ROS在中耳胆脂瘤的平均荧光值(112.41±14.61)明显高于其在正常皮肤的平均荧光值(15.23±1.97)(t=3.24,P<0.01);Western blot免疫印迹法检测显示,胆脂瘤中P-AKT的平均灰度值(0.95±0.08)明显高于正常皮肤中的平均灰度值(0.41±0.07)(t=2.04,P<0.05);在中耳胆脂瘤标本中,ROS与P-AKT的表达呈显著正相关(r=0.508,P<0.01)。结论中耳胆脂瘤组织中ROS和P-AKT均过度表达,明显高于正常皮肤组织,且两者呈正相关,ROS可能通过PI3K/AKT信号通路参与了中耳胆脂瘤的发病过程。
