老化型C57BL/6J小鼠耳蜗外侧壁血管纹Claudin-5、 NF-κB、P21的表达

Expression of Claudin-5, NF-κB and P21 in Stria Vascularis of the Lateral Wall of the Cochlea of Aging C57BL/6J Mice

摘要
目的探讨紧密连接蛋白(Claudin-5)、核转录因子(NF-κB)、细胞周期抑制蛋白(P21)在老化型C57BL/6J小鼠耳蜗外侧壁血管纹内的表达及意义。方法将C57BL/6J小鼠分为青年组(3月龄)和老年组(24月龄),每组各20只,检测两组小鼠听性脑干反应(ABR)的反应阈,分别行组织学切片观察耳蜗外侧壁血管纹形态结构,免疫组化检测Claudin-5、NF-κB、P21在血管纹表达,反转录聚合酶链反应(RT-PCR)检测Claudin-5、NF-κB、P21在耳蜗的表达水平。结果与青年组小鼠相比,老年组C57BL/6J小鼠ABR阈值升高,差异有统计学意义(均为P<0.05);老年组C57BL/6J小鼠血管纹内细胞上Claudin-5表达减少,NF-κB、P21表达增多,差异有统计学意义(均为P<0.01);RT-PCR检测显示老年组耳蜗内Claudin-5含量明显减少,NF-κB、P21含量明显增多,差异均有统计学意义(均为P<0.01)。结论 Claudin-5、 NF-κB、P21在老年组C57BL/6J小鼠耳蜗外侧壁血管纹内均有表达,Claudin-5表达降低可能与血管纹通透性相关,NF-κB和P21表达增多可能与内外淋巴循环紊乱和细胞凋亡有关。
