中耳胆脂瘤中DAPI核染色、FOXO3和Bim的表达和意义

Expression and Significance of DAPI Staining, FOXO3 and Bim in Middle Ear Cholesteatoma

摘要
目的检测中耳胆脂瘤中人类叉头框O3(forkhead box O3,FOXO3)、唯BH3域蛋白(Bcl-2 interacting mediator of cell death, Bim)的表达和相关性,以及Bim和4',6-二脒基-2-苯基吲哚(4',6-diamidino-2-phenylindole, DAPI)染色的相关性,探讨FOXO3和Bim在中耳胆脂瘤中的作用。方法应用免疫组化染色(immunohistochemistry)检测30例中耳胆脂瘤标本及15例正常皮肤中FOXO3、Bim的表达和定位;通过免疫荧光技术(immunofluorescence, IF)检测这些标本中Bim和DAPI的染色情况;应用Western blot检测以上标本FOXO3、Bim及内参的表达情况;分析Bim和DAPI核染色的相关性、Bim和FOXO3表达的相关性。结果 (1)免疫组化染色显示,FOXO3在胆脂瘤和正常皮肤的细胞核及胞质均有着色,在胆脂瘤细胞核中FOXO3的阳性样本率(36.7%,11/30)明显低于正常皮肤(86.7%,13/15)(χ2=10.04,P<0.05);胆脂瘤细胞胞质中FOXO3的阳性率(30.0%,10/30)也显明低于正常皮肤(80.0%,12/15)(χ2=10.04,P<0.05);胆脂瘤细胞胞质中FOXO3的阳性率(30.0%,10/30)也显明低于正常皮肤(80.0%,12/15)(χ2=8.72,P<0.05)。Bim主要定位于胆脂瘤和正常皮肤的细胞质,其在胆脂瘤中的阳性率(53.3%,16/30)明显低于正常皮肤组织(93.3%,14/15)(χ2=8.72,P<0.05)。Bim主要定位于胆脂瘤和正常皮肤的细胞质,其在胆脂瘤中的阳性率(53.3%,16/30)明显低于正常皮肤组织(93.3%,14/15)(χ2=7.20,P<0.05)。(2)免疫荧光染色显示DAPI和Bim在胆脂瘤的荧光值(分别为25.37±3.67和35.16±4.21)均弱于正常皮肤(分别为91.45±10.14和82.51±7.34),差异有统计学意义(t=1.93,P<0.05;t=1.71,P<0.05)。(3)Western blot检测显示:胆脂瘤中FOXO3和Bim的表达均明显低于正常皮肤,差异具有统计学意义(t=2.01,P<0.05;t=1.99,P<0.05)。(4)在中耳胆脂瘤标本中,FOXO3和Bim呈正相关(r=0.362,P<0.05);Bim和DAPI呈显著正相关(r=0.501,P<0.01)。结论 FOXO3/Bim信号通路在中耳胆脂瘤的形成过程中发挥了作用,且该作用与抗凋亡有关。
