水杨酸钠诱导耳鸣大鼠海马中TNF-α、IL-1β的表达

摘要
目的探讨水杨酸钠诱导耳鸣大鼠海马中肿瘤坏死因子α(TNF-α)、白介素1β(IL-1β)和N-甲基-D-天冬氨酸受体2B亚基(N-methyl-D-aspartate receptor 2B, NMDAR2B)的表达变化与大鼠耳鸣之间的联系。方法将18只健康成年SD大鼠随机分为三组:对照组(腹腔注射生理盐水)、腹腔注射水杨酸钠14天组(S14组)和腹腔注射水杨酸钠14天后停药恢复7天组(R7组),每组6只。检测各组大鼠造模前和造模结束后的惊跳反射前脉冲抑制(prepulse inhibition,PPI)和听性脑干反应(auditory brainstem response,ABR);检测后处死大鼠并迅速分离出海马组织,采用荧光定量PCR (quantitative PCR,qPCR)和Western Blot检测各组大鼠海马中TNF-α、IL-1β和NMDAR2B的表达。结果 (1)S14组大鼠前脉冲抑制率较对照组显著增加(P<0.001),提示耳鸣模型建立成功;(2)S14组大鼠的ABR反应阈为51±1.87 dB SPL,较对照组(36.67±1.05 dB SPL)显著升高(P<0.01);R7组大鼠的ABR反应阈为35±1.29 dB SPL,与对照组差异无统计学意义(P>0.05);(3)S14组大鼠海马TNF-α、IL-1β和NMDAR2B的表达水平较对照组显著升高(P<0.01);与对照组相比,R7组大鼠海马IL-1β的表达水平显著升高(P<0.05),TNF-α和NMDAR2B的表达水平与对照组相比差异无统计学意义(P>0.05)。结论腹腔注射水杨酸钠可诱导大鼠产生耳鸣,并可能通过上调大鼠海马区TNF-α、IL-1β和NMDAR2B的表达水平参与耳鸣的发生发展。
