阿司匹林联合中药复聪汤对老年性聋小鼠耳蜗外毛细胞存活及prestin蛋白表达的影响

摘要

目的探讨中西药联合应用上调耳蜗外毛细胞(outer hair cells, OHCs)prestin蛋白表达及促进耳蜗毛细胞存活的影响,为老年性聋的预防和治疗提供参考。方法选用4周龄DBA/2J小鼠120只,雌雄各半,分为六组:200 mg/kg阿司匹林加中药复聪汤治疗组、200 mg/kg阿司匹林治疗组、100 mg/kg阿司匹林加中药复聪汤治疗组、100 mg/kg阿司匹林治疗组、中药复聪汤治疗组和饮水对照组,每组20只。在试验前、治疗30、60和90天后检测各组动物ABR反应阈;在治疗90天后,处死动物,取出耳蜗,分别进行耳蜗全基底膜免疫组化prestin表达标记、硝酸银染色全耳蜗铺片毛细胞计数观察、扫描电镜观察和Western blot耳蜗蛋白印迹检测。结果实验前6组小鼠ABR反应阈(均为40～50 dB SPL)差异无统计学意义(P>0.05),治疗后,疗效最好的是200 mg/kg阿司匹林加中药复聪汤治疗组(治疗后30、60、90天ABR反应阈值分别为46.38±6.66、44.44±5.33、48.91±4.12 dB SPL),其次是单独中药复聪汤治疗组(治疗后30、60、90天ABR反应阈值分别为50.02±0.00、50.88±8.28、53.13±3.28 dB SPL),这两组治疗90天后与治疗前比较ABR反应阈差异无统计学意义(P>0.05),其余各组治疗90天后ABR反应阈较治疗前明显升高(P<0.05)。prestin蛋白表达强度在200 mg/kg阿司匹林加中药复聪汤治疗组最强,200 mg/kg阿司匹林治疗组渐弱,100 mg/kg阿司匹林加中药复聪汤治疗组最弱, 100 mg/kg阿司匹林治疗组、单独中药复聪汤治疗组和饮水对照组几乎没有表达。耳蜗铺片毛细胞计数和扫描电镜观察毛细胞的病变程度显示单独中药治疗组效果最好,其次是200 mg/kg阿司匹林加中药治疗组,这与ABR反应阈值和OHCs prestin表达强度不一致。结论 200 mg/kg阿司匹林联合中药复聪汤对老年性聋小鼠耳蜗OHCs prestin蛋白表达上调明显,对听功能和毛细胞有一定的保护作用。
