川芎嗪对长期注射水杨酸钠耳毒性拮抗作用的研究

摘要
目的探讨川芎嗪对长期注射水杨酸钠致听损伤的耳毒性拮抗作用。方法 50只健康级白色雄性SD大鼠随机分为生理盐水对照组(SC组,16只,每天腹腔注射生理盐水2.5 ml/kg)、水杨酸钠组(SS组,17只,每天腹腔注射400 mg/kg水杨酸钠溶液)和川芎嗪治疗组(TMP+SS组,17只,每天先腹腔注射400 mg/kg水杨酸钠溶液后,立即在对侧腹腔注射40 mg/kg盐酸川芎嗪溶液),各组连续注射14天后,在第15天比较各组体重、畸变产物耳声发射(DPOAE)幅值、听性脑干反应(ABR)阈值和潜伏期。结果第15天,SS组、TMP+SS组与SC组相比,体重明显减轻,差异有统计学意义(P<0.05);在f_2为1 000、2 000、3 000、4 000、6 000 Hz时,SS组DPOAE幅值较SC组升高(P<0.05);TMP+SS组与SC组相比,DPOAE幅值差异无统计学意义(P>0.05);在f_2为1 000、4 000、6 000 Hz时,SS组DPOAE幅值较TMP+SS组升高(P<0.05);SS组与SC组相比,ABR阈值升高,波Ⅱ潜伏期延长,差异有统计学意义(P<0.05),TMP+SS组与SC组比较,波Ⅱ潜伏期延长,差异有统计学意义(P<0.05)。结论川芎嗪对长期注射水杨酸钠致听损伤耳毒性的拮抗作用较弱,能部分拮抗长期注射水杨酸钠引起的DPOAE幅值增高,但对水杨酸钠导致的ABR阈值和潜伏期变化的拮抗作用还需要进一步研究。
